چکیده

Relaxin family peptide receptors (RXFP, nomenclature as agreed by the NC-IUPHAR Subcommittee on [18, 81]) may be divided into two pairs, RXFP1/2 and RXFP3/4. Endogenous agonists at these are heterodimeric hormones structurally related to insulin: relaxin-1, relaxin, relaxin-3 (also known INSL7), insulin-like 3 (INSL3) INSL5. Species homologues of relaxin have distinct pharmacology interacts with RXFP1, RXFP2 RXFP3, whereas mouse rat selectively bind activate RXFP1 [184]. is ligand for RXFP3 but it also binds RXFP4 has differential affinity between species [183]. INSL5 a weak antagonist RXFP3. INSL3 multiple complex binding interactions [189] [91] which direct N-terminal LDLa modules together linker domain act tethered receptor signaling [186]. interact their using residues in B-chains binding, activation, respectively [225, 104].

Relaxin family peptides and their receptors.

There are seven relaxin family peptides that are all structurally related to insulin. Relaxin has many roles in female and male reproduction, as a neuropeptide in the central nervous system, as a vasodilator and cardiac stimulant in the cardiovascular system, and as an antifibrotic agent. Insulin-like peptide-3 (INSL3) has clearly defined specialist roles in male and female reproduction, relaxi...

Relaxin-Family Peptide Receptors 1 and 2 Are Fully Functional in the Bovine

In most mammals the peptide hormone relaxin is a key physiological component regulating early pregnancy and birth. However, synteny analysis shows that the gene encoding ovarian relaxin-2 is deleted in cows and sheep. While, these ruminants appear to exhibit a relaxin-like physiology, as in other mammals, until now a molecular understanding of this has been lacking. Cloning and expression analy...

Relaxin family peptide receptors RXFP1 and RXFP2 modulate cAMP signaling by distinct mechanisms.

Two orphan leucine-rich repeat-containing G protein-coupled receptors were recently identified as targets for the relaxin family peptides relaxin and insulin-like peptide (INSL) 3. Human gene 2 relaxin is the cognate ligand for relaxin family peptide receptor (RXFP) 1, whereas INSL3 is the ligand for RXFP2. Constitutively active mutants of both receptors when expressed in human embryonic kidney...

The A-chain of human relaxin family peptides has distinct roles in the binding and activation of the different relaxin family peptide receptors.

The relaxin peptides are a family of hormones that share a structural fold characterized by two chains, A and B, that are cross-braced by three disulfide bonds. Relaxins signal through two different classes of G-protein-coupled receptors (GPCRs), leucine-rich repeat-containing GPCRs LGR7 and LGR8 together with GPCR135 and GPCR142, now referred to as the relaxin family peptide (RXFP) receptors 1...

H2 relaxin is a biased ligand relative to H3 relaxin at the relaxin family peptide receptor 3 (RXFP3).

Relaxin family peptide 3 receptors (RXFP3) are activated by H3-relaxin to inhibit forskolin-stimulated cAMP accumulation and stimulate extracellular signal-regulated kinase (ERK) 1/2 phosphorylation. In this study, we sought to identify novel signaling pathways coupled to RXFP3 and to investigate whether other members of the relaxin peptide family activated these pathways. Two patterns of signa...
